Well, it needs a graphics card, the 640x480 8bit Toby card is the classic.
Also, I recommend an Asante NB3 Ethernet, AUI/10base2/10baseT.

A IIcx is kind of old for a Jackhammer. I would probably go with OS 6.08, put in an old Supermac or Rasterops high end card (you know the ones you rarely see sold from the very early 90's full of chips), an old 50 pin SCSI card like a Micronet Nuport II, and to finish it off a rare specialty card like an early Audiomedia I or a Spigot.
One of my IIcx currently has a hardware SCSI card that mirrors the main HD and switches over incase of a fault. Have to finish setting that up.
I prefer cutting edge cards in quadras where the CPU complement the added speed of a good video card, scsi card, and maybe a DSP card.
The older 030's are fun to play with the earlier rare oddball cards.

"Must have" is pretty subjective. Obviously with that machine, video, and then ethernet, would be your highest priorities. But it depends what you want to do with it.
If you want to use Photoshop, then obviously you'll want to go for one of the higher-end DSP accelerated video cards, and fast SCSI. If you don't see yourself networking this machine much, you could use Appletalk when needed and free up another Nubus slot for something else. If you want to do any music work, then a better soundcard and fast SCSI would be a must. On the other hand if you want to do any analog to digital data capture, you could look for an old Nubus DAQ (Data AcQuisition card) from National Instruments or Hewlett Packard etc.
As far as soundcards go, the Digidesign Audiomedia I and II are pretty great, and cheap. The MediaVision ProAudio Spectrum 16 (Nubus or LC PDS card with breakout box) is really nifty, in that it gives you four stereo inputs (mixed by software-controlled hardware in the BoB down to one stereo A/D converter), MIDI in and out, and a headphone out, as well as a PC joystick port :?: for some reason. However the analogue converters on the Digis are much nicer, and the AM II includes digital audio I/O (RCA S/PDIF)
There's also the Digidesign SampleCell Nubus, which is a hardware sampler with 8 outs and a nice Mac GUI.
Course you could always save up several hundred dollars and try and score one of the Lexicon Nuverbs than come up for sale every, oh, two or three years [] ]'>

OOh! A Rocket in upgrade mode would be pretty hawt too.
If you use a Toby card, please also use the 13" fixed frequency monitor that goes with it, and use MaxAppleZoom or Monitor Expander to do the magic thing.
They can put a custom mode timing on the Toby card to fill the black monitor band with extra pixels!

Laboratory work. Measuring voltages coming from various sensors and graphing it over time

Do you use labview for that?
I have a couple cards, mostly just DIO and a GPIB card, I also have a NI DSP card (came with a video capture card oddly enough, not sure if they were used together). If I can find an analog I/O card I can do something interesting with them.
